Although more time consuming, dry loading of the sample provides the best chance of purity as it increases the resolution potential of the separation. PuriFlash Software provides two settings for dry loading: Standard and Auto. Standard equilibrates the column, pauses the pump, sample is injected on the column and then the pump resumes. In Auto mode, the equilibration step is skipped and the sample is loaded on to a dry column.

The dry load column is placed on top of the purification column and the pump resumes. Auto does not equilibrate the purification column. The dry load column is placed on top of the purification column and the pump is activated.
